AbstractHigh molecular weight PEI (600 K) modified multiwalled carbon nanotubes (HMWPEI-MWCNTs) were studied in 293T cells as safe and efficient gene delivery vehicles. The HMWPEI-MWCNTs exhibited low cytotoxicity, and had enough ability to condense DNA and obviously enhance gene transfer activity. The concentration of HMWPEI-MWCNTs to 100 mu g/mL showed less effect on the 60% of cell viability, while the cell viability of HMWPEI was around 20% at the same concentration. The transfection efficiency of HMWPEI-MWCNTs was around 1.8 times, and much higher than those of naked DNA under an optimal nitrogen/phosphate (N/P) ratio of 10:1. The study demonstrates the possibility of developing the novel safe and efficient gene delivery systems by adopting low toxic and biocompatible nanocomposite.
