激光诱变筛选木霉菌株及木聚糖酶发酵条件优化

Abstract本实验利用激光这一高效诱变手段,对野生菌株绿色木霉Tr-02进行辐照诱变,通过两轮的定向筛选,最终得到一株高产木聚糖酶的菌株。对突变株进行发酵条件优化,结果表明,以0.5%麸皮为碳源,0.5%蛋白胨为氮源,0.1% Tween-80作为产酶促进剂,培养基初始pH值为5.0,瓶装量100mL,接种两环,28℃,180r/min培养48h,木聚糖酶活力达到281.21U·mL-1,较之相同条件出发菌株木聚糖酶活力增长31.83%。
